Novel Antisense Therapy Bepirovirsen Advances Hepatitis B Treatment Options

A phase 3 clinical trial of bepirovirsen, an antisense oligonucleotide therapy, has demonstrated promising results for chronic hepatitis B treatment, with findings published in The New England Journal of Medicine. This breakthrough represents a significant advancement in addressing the treatment needs of approximately 296 million people living with chronic hepatitis B worldwide.

Unlike conventional nucleoside analogue therapies that primarily suppress viral replication, bepirovirsen employs a novel mechanism targeting hepatitis B virus RNA to reduce surface antigen production. The trial evaluated the therapy’s efficacy through primary endpoints including sustained HBsAg loss and undetectable HBV DNA levels, metrics aligned with WHO definitions of functional cure.

This novel approach addresses a critical limitation of current standard care, which rarely achieves HBsAg loss despite effective viral suppression. The positive trial results suggest bepirovirsen could transform the hepatitis B treatment landscape by offering patients improved prospects for functional cure beyond traditional viral suppression strategies.
