Two phase III randomised controlled trials presented at the American Diabetes Association conference in June 2026 demonstrate that orforglipron, a once-weekly GLP-1 receptor agonist, delivers clinically significant HbA1c reductions when combined with metformin or insulin glargine. Two parallel phase III randomised controlled trials published in The Lancet and JAMA (June 2026) present compelling efficacy data for orforglipron in type 2 diabetes management. When combined with metformin, orforglipron achieved a mean HbA1c reduction of 1.5%, while the combination with insulin glargine produced a 1.2% reduction from baseline. These reductions exceed the GLP-1 class mean of 1.0% observed in monotherapy trials, suggesting additive benefits in combination settings. The trials enrolled patients with type 2 diabetes on standard-of-care therapies, enhancing real-world applicability. Beyond efficacy, the once-weekly dosing schedule addresses a key barrier to medication adherence in diabetes care. Results were presented at the American Diabetes Association annual meeting in June 2026.
