Two parallel phase III randomised controlled trials published in The Lancet and JAMA (June 2026) present compelling efficacy data for orforglipron in type 2 diabetes management. When combined with metformin, orforglipron achieved a mean HbA1c reduction of 1.5%, while the combination with insulin glargine produced a 1.2% reduction from baseline. These reductions exceed the GLP-1 class mean of 1.0% observed in monotherapy trials, suggesting additive benefits in combination settings. The trials enrolled patients with type 2 diabetes on standard-of-care therapies, enhancing real-world applicability. Beyond efficacy, the once-weekly dosing schedule addresses a key barrier to medication adherence in diabetes care. Results were presented at the American Diabetes Association annual meeting in June 2026.
