A 2020 systematic review in Nutrients documents vitamin D deficiency as a significant risk factor across rickets, osteoporosis, multiple sclerosis, type 1 diabetes, inflammatory bowel disease, and increased infection susceptibility. The comprehensive analysis by Charoenngam and Holick from Boston University School of Medicine reveals how suboptimal vitamin D status impairs immune regulation, creating vulnerability to both infectious and autoimmune conditions.

The research distinguishes between direct causative mechanisms—such as vitamin D’s essential role in calcium homeostasis for bone health—and indirect associations mediated through immune dysfunction. This distinction is critical for understanding clinical implications and informing prevention strategies. The bidirectional nature of vitamin D and disease suggests that monitoring status becomes particularly important in patients with chronic conditions.
