New clinical data from the National Institutes of Health reveals that 47% of patients experience significant weight-loss plateau within 12 months of beginning GLP-1 therapy with medications like Ozempic and Wegovy. This striking statistic underscores a critical challenge in obesity treatment and highlights the need for better understanding of medication resistance mechanisms.

The NIH research team identified receptor desensitization as the primary biological cause, occurring in approximately 40% of the patient population. Brain imaging studies demonstrated that while initial weight loss is promising—with 89% of patients maintaining losses at three months—effectiveness steadily declines, dropping to 63% by month 12 and 53% by month 18.

These findings suggest that individual genetic and neurobiological differences significantly influence treatment outcomes. The variability in brain cell responses to semaglutide underscores the importance of personalized medicine approaches in managing obesity and developing strategies to prevent or overcome treatment plateaus.
