Zanidatamab Emerges as Promising Alternative to Standard HER2-Targeted Therapy in Gastric Cancer

A groundbreaking phase III trial published in The New England Journal of Medicine has demonstrated that zanidatamab, a novel bispecific antibody, significantly outperforms the current standard of care in treating HER2-positive gastroesophageal cancer. The HERIZON-GEA-01 trial enrolled 710 patients across multiple international sites and revealed that zanidatamab plus chemotherapy reduced the risk of disease progression or death by 44% compared to trastuzumab-based treatment. The bispecific antibody works by simultaneously targeting two distinct epitopes on the HER2 receptor, a mechanism that investigators believe enhances anti-tumor activity. Median progression-free survival reached 12.5 months with zanidatamab versus 8.5 months with trastuzumab. The drug demonstrated a manageable safety profile, positioning it as a potential treatment option for thousands of patients with this aggressive cancer type globally.
