Mitochondria have long been understood as cellular powerhouses, but emerging research reveals a far more sophisticated role. Recent biochemical studies demonstrate that tricarboxylic acid (TCA) cycle intermediates function as master signalling molecules, orchestrating immune responses, stem cell maintenance, and tumour development independent of ATP production.

Key metabolites including acetyl-CoA, α-ketoglutarate, itaconate, succinate, and fumarate operate as pleiotropic regulators with profound epigenetic consequences. Acetyl-CoA drives histone acetylation and lymphatic vessel formation, while α-ketoglutarate balances stem cell preservation with immune activation. Itaconate emerges as a critical immunometabolite with anti-inflammatory properties, whereas succinate and fumarate accumulation can promote oncogenic epigenetic dysregulation.

This mechanistic understanding reshapes therapeutic approaches. Supporting mitochondrial health through aerobic exercise, calorie restriction, and targeted micronutrients—including B vitamins, magnesium, CoQ10, and alpha-lipoic acid—optimizes TCA cycle flux and downstream cellular signalling pathways.
