What is 7-Keto DHEA?

7-Keto DHEA (3-acetyl-7-oxo-dehydroepiandrosterone) is a metabolite of dehydroepiandrosterone (DHEA), a hormone naturally produced by the adrenal glands. Unlike DHEA, 7-Keto DHEA does not convert into sex hormones like testosterone or estrogen, making it an attractive alternative for those seeking DHEA’s potential benefits without hormonal effects. This synthetic compound is primarily marketed as a weight loss supplement and anti-aging product, though scientific evidence for these claims remains limited.

7-Keto DHEA was first identified in the 1990s and has since been studied for its potential effects on metabolism, immune function, and body composition. The compound is heat-stable and does not undergo conversion to androgens or estrogens in the body, distinguishing it from its parent compound DHEA.

Health benefits & uses

Weight loss and metabolism (Limited evidence): Small studies suggest 7-Keto DHEA may support modest weight loss when combined with diet and exercise. One randomized controlled trial found participants taking 200mg daily lost more weight than placebo groups, but the study was small and short-term.

Immune function support (Limited evidence): Some research indicates 7-Keto DHEA may help maintain immune function in older adults, potentially reducing age-related immune decline. However, studies are preliminary and require further validation.

Anti-aging effects (Limited evidence): Proponents claim 7-Keto DHEA may help combat age-related metabolic slowdown, but robust clinical evidence supporting significant anti-aging benefits is lacking.

Body composition (Limited evidence): Some studies suggest potential improvements in lean body mass, though results are inconsistent and based on small sample sizes.

How it works

7-Keto DHEA is thought to work by activating thermogenic enzymes in the liver, potentially increasing metabolic rate and energy expenditure. The compound may influence the activity of three thermogenic enzymes: fatty acid CoA oxidase, malic enzyme, and glycerol-3-phosphate dehydrogenase. These enzymes play roles in fat oxidation and cellular energy production.

Unlike DHEA, 7-Keto DHEA does not bind to androgen or estrogen receptors, avoiding hormonal side effects. Instead, it appears to work through metabolic pathways that may enhance the body’s ability to burn calories and maintain lean muscle mass. The compound may also support immune function through mechanisms that are not yet fully understood.

Dosage

Typical dosing ranges for 7-Keto DHEA supplements are 100-200mg daily, usually divided into two doses taken with meals. Most studies have used doses between 100-200mg per day. There is no established Recommended Dietary Allowance (RDA) or Upper Limit (UL) for 7-Keto DHEA, as it is not considered an essential nutrient.

Due to limited long-term safety data, it is recommended to consult with a healthcare provider before beginning supplementation, especially for extended periods. Some practitioners recommend starting with lower doses (50-100mg) to assess tolerance before increasing to higher amounts.

Food sources

7-Keto DHEA is not naturally present in foods in meaningful amounts. While DHEA is produced naturally by the human body and found in trace amounts in some foods, 7-Keto DHEA as used in supplements is synthetically produced. The body does produce small amounts of 7-Keto DHEA as a natural metabolite of DHEA, but supplemental forms provide much higher concentrations than what occurs naturally.

Safety & side effects

7-Keto DHEA is generally well-tolerated in healthy adults when used short-term. Reported side effects are typically mild and may include nausea, dizziness, low blood pressure, and fatigue. Some users report heartburn or metallic taste.

Long-term safety data is limited, and the effects of prolonged use are not well-established. Unlike DHEA, 7-Keto DHEA does not appear to cause hormonal side effects such as acne, hair loss, or voice changes, since it does not convert to sex hormones.

The supplement may affect thyroid hormone levels in some individuals, though clinical significance remains unclear. Regular monitoring may be advisable for those using the supplement long-term.

Interactions

Limited data exists on drug interactions with 7-Keto DHEA. Potential interactions may include:

Diabetes medications: May affect blood sugar levels, potentially requiring dosage adjustments of diabetes medications.

Thyroid medications: Some evidence suggests 7-Keto DHEA may influence thyroid hormone levels, potentially affecting thyroid medication effectiveness.

Other supplements: Effects of combining 7-Keto DHEA with other metabolic supplements or hormone-affecting compounds are not well-studied.

Who should be cautious

Certain groups should exercise particular caution or avoid 7-Keto DHEA:

Pregnant and breastfeeding women: Safety during pregnancy and lactation has not been established.

Individuals with thyroid disorders: The supplement may affect thyroid function and could interfere with thyroid medications.

People with diabetes: May affect blood glucose levels and require monitoring.

Those with liver conditions: Limited data on safety in individuals with liver disease.

Children and adolescents: Safety and efficacy have not been established in younger populations.
