What is MK-677 / Ibutamoren?

MK-677, also known as Ibutamoren or LUM-201, is a selective ghrelin receptor agonist and growth hormone secretagogue. Originally developed by Reverse Pharmacology for treating growth hormone deficiency, this compound mimics the action of ghrelin, the “hunger hormone,” to stimulate the release of growth hormone and insulin-like growth factor 1 (IGF-1) from the pituitary gland.

Unlike anabolic steroids or direct growth hormone injections, MK-677 works by activating the body’s natural growth hormone production pathways. It is administered orally and has a longer half-life than natural ghrelin, making it more practical for therapeutic applications. Currently, MK-677 is not approved by the FDA for general use and remains an investigational compound being studied for various medical conditions.

Health benefits & uses

Muscle mass and bone density (Moderate evidence): Clinical trials have demonstrated that MK-677 can increase lean body mass and bone mineral density in elderly adults. A 12-month study showed significant improvements in bone turnover markers and modest increases in muscle mass, though functional strength improvements were limited.

Sleep quality (Limited evidence): Some research suggests MK-677 may improve sleep quality by increasing REM sleep duration. However, studies are small and more research is needed to confirm these effects.

Growth hormone deficiency (Limited evidence): While MK-677 effectively raises growth hormone and IGF-1 levels, its clinical efficacy for treating diagnosed growth hormone deficiency requires further study compared to established treatments.

Anti-aging effects (Limited evidence): Though marketed for anti-aging benefits, robust clinical evidence supporting these claims is lacking. Most studies have been conducted in elderly populations or those with specific deficiencies.

How it works

MK-677 functions as a ghrelin receptor agonist, binding to ghrelin receptors (GHSR) in the brain and pituitary gland. This binding triggers a cascade of events that stimulates the release of growth hormone from somatotroph cells in the anterior pituitary. The increased growth hormone then stimulates the liver to produce more IGF-1, which mediates many of growth hormone’s anabolic effects.

Unlike exogenous growth hormone administration, MK-677 preserves the natural pulsatile pattern of growth hormone release, which may result in fewer side effects. The compound also increases appetite through its action on ghrelin receptors, which can be beneficial for individuals with poor appetite or muscle wasting conditions.

Dosage

Since MK-677 is not approved for general use, there are no established recommended dosages. Research studies have typically used doses ranging from 10-25 mg daily, taken orally. Most clinical trials have used 25 mg once daily, often administered in the evening due to its potential to improve sleep quality and mimic natural growth hormone release patterns.

Given the experimental nature of this compound, any use should only be considered under strict medical supervision as part of a clinical trial or approved research protocol. Self-medication with MK-677 is not recommended due to potential risks and unknown long-term effects.

Food sources

MK-677 is a synthetic compound and does not occur naturally in foods. It is only available as a pharmaceutical compound produced in laboratories. Natural growth hormone production can be supported through adequate sleep, regular exercise, protein intake, and maintaining healthy body composition, but these methods do not replicate the specific effects of MK-677.

Safety & side effects

Clinical studies have reported several side effects associated with MK-677 use. Common adverse effects include increased appetite, mild swelling (edema), muscle pain, and elevated blood glucose levels. Some participants have experienced increased insulin resistance, which is concerning for individuals at risk of diabetes.

Other reported side effects include fatigue, joint pain, and in some cases, elevated cortisol levels. Long-term safety data is limited, and the compound’s effects on cancer risk remain unclear, as growth hormone and IGF-1 can potentially promote tumor growth in predisposed individuals.

Interactions

Limited data exists regarding drug interactions with MK-677. However, given its effects on glucose metabolism and insulin sensitivity, it may interact with diabetes medications, potentially altering blood sugar control. The compound might also interact with medications that affect growth hormone or IGF-1 levels.

Caution is advised when combining MK-677 with other supplements or compounds that affect growth hormone, insulin sensitivity, or appetite regulation. Always consult healthcare providers about potential interactions before considering use.

Who should be cautious

Several groups should avoid MK-677 or use it only under strict medical supervision. Individuals with diabetes or prediabetes should be particularly cautious due to the compound’s effects on glucose metabolism and insulin sensitivity.

People with a history of cancer should avoid MK-677, as elevated growth hormone and IGF-1 levels may potentially promote tumor growth. Pregnant or breastfeeding women should not use this compound due to unknown effects on fetal development.

Individuals with heart conditions, liver disease, or kidney problems should also exercise extreme caution, as should anyone taking medications for chronic conditions without thorough medical evaluation.
